Purification and Identification of Falcarinol, Polyacetylene Family, from Glehnia littoralis Capable of Cytotoxicity on Jurkat T Lymphoma

Abstract
A cytotoxic material on human acute lymphoma jurkat T cells was identified with dichloromethaneextracts from root of Glehnia litoralis. The extract was further purified through silica gel column chromatography and preparative HPLC 18 chromatography. The purified cytotoxic substance of Glehnia littoralis was estimated to have about 98% purity by GC/Mass analyzer and designated ¡°BPCH2¡±. Chemical structure of the BPCH2 was analyzed with GC/Mass, 1H-NMR, 13C-NMR, DEPT, HMQC, and HMBC spectrum. BPCH2 is identified as (Z)heptadeca-1,9-diene-4,6-diin-3-ol (falcarinol) its molecular formula is C17H24O and its molecular weight is 244Da. Falcarinol purification method from Glehnia littoralis is established and the maferial has strong cytotoxicity. This material may use anti-tumor agent, health promotion agent or tonic material.
